Electron Microscopic Observations on Lymph Node Cells Stimulated by Antigenic Substance

Abstract
The aim of the present study was to determine the nature of the stimulated lymph node cells following the subcutaneous injection of soluble antigen.
After subcutaneous injection of bovine gamma globulin or emulsion of boyine gamma globulin and incomplete Freund¢¥s adjuvant into the dorsal foot-pad of the rabbit, the draining popliteal lymph node was removed 3, 7, 10, 13, and 20 days after injection respectively. The lymph nodes were examined histologically with hematoxylineosin and methylgreen-pyronin stainings and stimulated lymph node cells were examined by electron microscope. And following conclusions were obtained.
1. The lymph node stimulated by soluble antigen revealed marked proliferative responses chiefly of "large pyroninophilic cells". The proliferative responses were most evident in the secondary nodule in a few days after antigenic stimulation, and there after in the peripheral areas of primary nodule and in the medulary cord.
2. The proliferated cells thus examined by electron microscope were found to fall into two classes, according to the current morphologic terminology; some were in the category of the lymphocytes and others, the category of the plasma cells.
3. The proliferated cells, both lymphocytes and plasma cells, following antigenic stimulation were thought to produce antibodies.
